Specificity

Clone 1N7 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects survivin. It targets an epitope within 21 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the internal region of human Survivin.

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Survivin in Jurkat cell lysate.

Tested applications

Affinity Binding Assay: A representative lot of this antibody bound Survivin peptide with a KD of 1.7 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Survivin in MCF-7 cells.

Target description

Baculoviral IAP repeat-containing protein 5 (UniProt: O15392; also known as Apoptosis inhibitor 4, Apoptosis inhibitor survivin) is encoded by the BIRC5 (also known as API4, IAP4) gene (Gene ID: 332) in human. Survivin, a multi-tasking protein, is the smallest member of the inhibitor of apoptosis (IAP) family of proteins that inhibits apoptosis by blocking the activity of caspases. It is also involved in the regulation of cell cycle. Overexpression of survivin is reported in a large number of malignancies. Survivin is expressed highly at G2/M phase and declines rapidly in G1 phase of cell cycle. It is a component of the chromosome passage protein complex (CPC) that is shown to be essential for chromosome alignment and segregation during mitosis and cytokinesis. It acts as an important regulator of the localization of the CPC complex by directing its movement to different locations from the inner centromere during prometaphase to midbody during cytokinesis. It also participates in the organization of the center spindle by associating with polymerized microtubules. Survivin can undergo phosphorylation at multiple sites. Phosphorylation at threonine 117 by Aurora kinase B that prevents its interaction with inner centromere protein and localization to mitotic chromosomes. Phosphorylation at threonine 48 by casein kinase 2 is shown to be important for its mitotic and anti-apoptotic activities. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.(Ref.: Jaiswal, PK., et al. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
