Specificity

Clone 1J2 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects C-Reactive Protein (CRP). It targets an epitope within 17 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the internal region of human C-Reactive Protein.

Application

Quality Control Testing

Evaluated by Western Blotting in Rat Kidney tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected C-Reactive Protein in Rat kidney tissue lysate.

Tested applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected C-Reactive Protein in rat kidney tissue sections.

Enzyme Immunoassay (ELISA) Analysis: A Serial of dilutions from a representative lot detected C-Reactive Protein in C-Reactive and GST Proteins.

Target description

C-reactive protein (UniProt: P02741; also known as CRP) is encoded by the CRP (also known as PTX1) gene (Gene ID: 1401) in human. CRP is an acute-phase protein that is synthesized by hepatocytes in response to pro-inflammatory cytokines during inflammation and infection. It is an evolutionarily conserved major acute-phase protein whose levels may increase over a 1,000-fold in severe inflammatory state. It is synthesized with a signal peptide (aa 1-18) that is subsequently cleaved off to produce the mature protein. Human CRP is a pentameric protein composed of five identical non-covalently bound subunits that bind to phosphocholine (PCh) in a Ca2+-dependent manner and leading to the activation of the complement system via the C1Q complex. Each subunit has one PCh-binding site and each subunit binds two calcium ions. Interleukin-1 and 6, produced in response to injury or infection, are two major triggers for its synthesis. In mature CRP, glutamic acid 81 in the PCh-binding site is reported to interact with the nitrogen atom of choline in PCh and phenylalanine 66 interacts with three methyl groups of choline. The phosphate group of PCh directly coordinates with the two Ca2+ bound to CRP. Under acidic pH conditions, native pentameric CRP can be transformed into a non-native CRP, which exposes a hidden ligand-binding site for non-PCh ligands and enables CRP to bind to immobilized, denatured, and aggregated proteins. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
