Specificity

Clone 3I16 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects DEAH box protein 37 (DHX37). It targets an epitope within 24 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal region of human DEAH box protein 37 (DHX37).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected DHX37 in HeLa cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected DHX37 in U2OS and Jurkat cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound DHX37 with a KD of 2.3 x 10-10 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected DHX37 in human placenta and human skin tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected DHX37 in MCF-7 cells.

Anti-DHX37, clone 3I16 ZooMAb®, Cat. No. ZRB1524, is a recombinant Rabbit monoclonal antibody that targets DHX37 and is tested for use in is used in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Probable ATP-dependent RNA helicase DHX37 (UniProt: Q8IY37; also known as EC:3.6.4.13, DEAH box protein 37) is encoded by the DHX37 (also known as DDX37, KIAA1517) gene (Gene ID: 57647) in human. DHX37, a member of the DEAD box helicase family, is an ATP-binding RNA helicase that plays a role in maturation of the small ribosomal subunit in ribosome biogenesis. It is required for the release of the U3 snoRNP from pre-ribosomal particles. Lack of DHX37 is reported to trigger a surveillance pathway that leads to degradation of pre-ribosomal particles and a depletion of DHX37 is shown to inhibit the conversion of the 21S pre-rRNA to 18SE. Its expression has been reported in fallopian tube, ovary, uterus, and testis and is also detected in the brain. Its DEAH box motif is localized to amino acids 372-375. It plays a role in early testis development and its pathogenic variants are reported to cause 46,XY gonadal dysgenesis and lead to testicular regression syndrome. Mutations in DHX37 gene are also reported to cause neurodevelopmental disorders with brain anomalies that are characterized by severe developmental delays, cortical volume loss and cerebellar hypoplasia. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
