Specificity

Clone 2B5 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Neuropeptide Y.

Application

Quality Control Testing

Evaluated by Immunohistochemistry (Paraffin) in human cerebral cortex tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution of this antibody detected Neuropeptide Y in human cerebral cortex tissue sections.

Tested applications

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Neuropeptide Y in SH-SY5Y cells.

Affinity Binding Assay: A representative lot of this antibody bound Neuropeptide Y with a KD of 5.8 x 10-8 in an affinity binding assay.

Target description

Pro-neuropeptide Y (UniProt: P01303) is encoded by the NPY gene (Gene ID: 4852) in human. Neuropeptide Y is one of the most abundant peptides in the nervous system and is also found in some chromaffin cells of the adrenal medulla. In the brain it is present in the cortex, hippocampus, hindbrain and hypothalamus. It is implicated in the control of feeding and in secretion of gonadotrophin-release hormone. It is shown to be one the most potent orexigenic peptides that stimulates food intake with a preferential effect on carbohydrate intake. It decreases latency to eat, increases motivation to eat and delays satiety by augmenting meal size. Its effects on feeding are mediated through the Y1 and Y5 receptors. Neuropeptide Y also acts as a vasoconstricting peptide that is released from sympathetic nerves in the heart and affects coronary artery constriction and myocardial contraction. It is also reported to potentiate the vasoconstrictor effects of other extracellular messengers, including a-adrenergic agonists and angiotensin II. It is also linked to inhibition of acetylcholine release from parasympathetic nerve endings in the heart. Neuropeptide Y is synthesized as a pre-propeptide with a signal peptide (aa 1-28) and a C-flanking peptide (aa 68-97) that are subsequently cleaved off to produce the active peptide (aa 29-64). This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
