Specificity
Clone 1G4 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects T-cell factor 4 (TCF-4). It targets an epitope within 20 amino acids from the N-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 20 amino acids from the N-terminal region of human T-cell factor 4 (TCF-4).
Application
Quality Control Testing
Evaluated by Western Blotting in Rat Liver tissue lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected TCF-4 in Rat Liver tissue lysate.
Tested applications
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TCF-4 in HeLa cells and A431 cells.
ELISA Analysis: A r representative lot of this antibody detected TCF-4 peptide in ELISA application..

Target description
Transcription factor 7-like 2 (UniProt: Q9NQB0; also known as HMG box transcription factor 4, T-cell-specific transcription factor 4, T-cell factor 4, TCF-4, hTCF-4) is encoded by the TCF7L2 (also known as TCF4) gene (Gene ID: 6934) in human. The mammalian T-cell factor family consists of four members (TCF1 4) that contain a highly conserved high-mobility-group (HMG) domain that is responsible for their binding to DNA. TCF-4 is the most prominent member of this family that is highly expressed in small intestine with highest expression at the top of crypts and a gradient expression from crypt to villus. It is also detected in colon epithelium and colon cancer and in mammary gland carcinoma. However, its expression in fetal small intestine is barely detectable. HMG box of TCF-4 is localized to amino acids 350-418. TCF-4 participates in the Wnt signaling pathway and modulates Myc expression by binding to its promoter in a sequence-specific manner. It acts as a repressor in the absence of catenin b1, and as an activator in its presence. It is shown to activate transcription from promoters with several copies of the Tcf motif 5′-CCTTTGATC-3′ in the presence of catenin b1. Transducin-like enhancer of split 1,2,3, and 4 (TLE1-4) are reported to repress transactivation mediated by TCF-4 and catenin b1. TCF-4 undergoes phosphorylation at Thr 201 or Thr 212 by Nemo-like kinase (NLK), which inhibits its DNA-binding and prevents transcriptional activation of target genes of the canonical Wnt/b-catenin signaling pathway. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
