Specificity
Clone 3N18 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Collagen Type III. It targets an epitope with 22 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal region of human Collagen a1(III) chain.
Application
Quality Control Testing
Evaluated by Western Blotting in HT1080 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Collagen Type III in HT1080 cell lysate.
Tested applications
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Collagen Type III in human skin tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound Collagen Type III with a KD of 6.7 x 10-6 in an affinity binding assay.
ELISA Analysis: 300 μg/mL from a representative lot detected Collagen Type III in Collagen Type III in an indirect ELISA.

Target description
Collagen alpha-1(III) chain (UniProt: P02461; also known as Collagen type IIIa) is encoded by the COL3A1 gene (Gene ID: 1281) in human. Collagen is the most abundant protein in the body that is predominantly synthesized by fibroblasts. However, epithelial cells may also synthesize small amounts of collagen. The collagen super family includes over 20 different types of collagen with at least 38 distinct polypeptide chains. Collagen type III occurs in most soft connective tissues along with type I collagen. It is a major structural component in hollow organs such as large blood vessels and uterus that must withstand stretching. Collagen type III is also involved in regulation of cortical development. It is reported to be the major ligand of ADGRG1 in the developing brain and binding to ADGRG1 inhibits neuronal migration and activates the RhoA pathway by coupling ADGRG1 to GNA13 and possibly GNA12. It is synthesized with a signal peptide (aa 1-23), an N-terminal propeptide (aa 24-153), and a C-terminal propeptide (aa 1222-1466), which are subsequently cleaved off to generate the mature form. Three identical type III procollagen chains come together in the C-terminal ends to form collagen III molecule and it is stabilized by the formation of disulfide bonds. Each individual chain folds into left-handed helix and the three chains are then wrapped together into a right-handed superhelix, the triple helix. Mutations in COL3A1 are linked to Ehlers-Danlos syndrome, a connective tissue disorder that is characterized by skin hyperextensibility, articular hypermobility, and tissue fragility. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
