Specificity

Clone 1I14 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Brain-derived neurotrophic factor (BDNF). It targets an epitope within 17 amino acids from the C-terminal half. Detects the pro- and the mature form of BDNF and is predicted to react with all isoforms.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal half of human Brain-derived neurotrophic factor (BDNF).

Application

Quality Control Testing

Evaluated by Western Blotting in Neuro2a cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected BDNF in Neuro2a cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected BDNF in human brain, mouse brain, rat brain, and PC12 cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected BDNF in Neuro2A cells.

Affinity Binding Assay: A representative lot of this antibody bound BDNF with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected BDNF in human cerebral cortex tissue sections.

Anti-BDNF, clone 1I14 ZooMAb®, Cat. No. ZRB1136, is a recombinant Rabbit monoclonal antibody that specifically targets BDNF and is tested for use in Affinity Binding Assay, Immunocytochemistry, , Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Brain-derived neurotrophic factor (UniProt: P23560; also known as BDNF, Abrineurin) is encoded by the BDNF gene (Gene ID: 627) in human. BDNF is a member of the neutrophin family that is synthesized with a signal peptide (aa 1-18) and a propeptide (aa 19-128). The Signal peptide is cleaved off to generate the proBDNF (aa 19-247) that is further proteolytically processed to produce the mature BDNF (aa 129-247). BDNF is an important signaling molecule that activates signaling cascades downstream of Neurotrophic Receptor Tyrosine Kinase 2 (NTRK2). The expression of BDNF is highly regulated and displays great variability in its levels in healthy subjects. Changes in BDNF expression are reported in both normal and pathological aging. It plays a key role in in survival and differentiation of neuronal populations during development and participates in axonal growth, pathfinding and in the modulation of dendritic growth and morphology. In the adult brain, it maintains high expression levels and regulates both excitatory and inhibitory synaptic transmission and activity-dependent plasticity. BDNF and proBDNF are shown to have opposing effects on cellular function. The proBDNF form is secreted under both pathological and non-pathological conditions and preferentially binds p75 NTR receptor, which facilitates long-term depression (LTD). On the other hand, mature BDNF binds specifically to tyrosine kinase receptors (TrkB) and promotes cell survival and facilitates Long-term potentiation (LTP). This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
