Specificity

Clone 2N18 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects human Proteasome subunit beta type-2 (PSMB2).

Immunogen

His-tagged full-length recombinant human Proteasome subunit beta type-2 (PSMB2).

Application

Anti-PSMB2, clone 2N18 ZooMAb, Cat. No. ZRB1059, is a recombinant Rabbit monoclonal antibody that specifically targets PSMB2 and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected PSMB2 in Jurkat and K562 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound recombinant PSMB2 protein with a KD of 3.1 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PSMB2 in human small intestine tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PSMB2 in HeLa, HUVEC, and A431 cells.

Target description

Proteasome subunit b type-2 (P49721; also known as EC:3.4.25.1, Macropain subunit C7-I, Multicatalytic endopeptidase complex subunit C7-I, Proteasome component C7-I) is encoded by the PSMB2 gene (Gene ID: 5690) in human. Proteasomes are large multi-subunit protease complexes, localized in the nucleus and cytosol that selectively degrade intracellular proteins. Proteasome consists of a multi-component enzymatic complex known as the 26S proteasome, which plays a key role in the maintenance of protein homeostasis by removing misfolded or damaged proteins that could impair cellular functions, and by removing proteins whose functions are no longer required. The proteolytic core of this complex, the 20S proteasome, contains multiple peptidase activities and functions as the catalytic machine. Unlike other proteases, the 20S proteasome ensures that virtually all peptide bonds within a protein substrate are susceptible to cleavage, by possessing multiple proteolytic activities in one proteolytic internal chamber. This core is composed of 28 subunits arranged in four heptameric, tightly stacked, rings ( 7, β7, β7, 7) to form a cylindrical structure. The proteolytic activity is confined to the β1, β2, and β5-subunits. The proteolytic sites are formed by N-terminal threonine residues that face the central cavity of the 20S complex. The regulatory unit of the 26S proteasome is known as the 19S (PA 700) particle and consists of at least 15 subunits that include ATPases, a de-ubiquitinating enzyme, and polyubiquitin-binding subunits. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
