Quality Control Testing
Evaluated by Dot Blot with ssDNA from Calf thymus.
Dot Blot Analysis: A 1:1,000 dilution of this antibody detected ssDNA from Calf thymus.
Tested applications
Enzyme Immunoassay Analysis (ELISA): Serial of dilutions from a representative lot detected ssDNA from Calf thymus.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ssDNA in Molt 4 cells treated with Etoposide (10 M; overnight).

Target description
DNA, the carrier of genetic information, is a crucial part of basic biological research and synthetic biology. It exists as a double-helix structure and the two DNA strands known as polypeptides are composed of monomeric units known as nucleotides. Each nucleotide consists of four bases: adenine, guanine, cytosine, thymine and a deoxyribose sugar moiety. The nitrogenous bases of the two separate strands are bound together by base pairing (A-T and C-G) to create a double stranded DNA. Under certain conditions, these two strands can come apart (melting) to generate two single-stranded DNA. Single-stranded DNA (ssDNA) is a crucial DNA damage signal found at the sites of DNA damage that helps in recruitment of checkpoint kinases to the site of damage. This leads to cell cycle arrest, transcriptional upregulation of DNA repair genes, and modulation of DNA replication pathways. Hence, ssDNA is considered as a key intermediate in nearly all biochemical reactions involving DNA replication and DNA repair. However, the ssDNA structure is not stable and single-stranded DNA-binding proteins (SSB) hold the ssDNA intact and exposed during the course of its function, such as DNA transcription, recombination, and repair, and serve as template for opposite strand DNA synthesis. Antibodies against ssDNA are commonly used in ELISA-based test for the quantitative determination of IgG antibodies against ssDNA and have been used in serological detection of systemic lupus erythematosus (SLE) and drug induced lupus. Physical form
Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
