General description

2xYT (2x Yeast Extract Tryptone medium) is a standard microbial growth medium used for the cultivation of E. coli and M13 bacteriophage. This nutrient-rich microbial broth contains peptides, amino acids, and water-soluble vitamins in a low-salt formulation.

Application

2x YT medium has been used for culturing E. coli BL21 for cloning , E. coli SS320 cells for scaffold synthesis , and NEB 5-alpha competent E. coli cells for protospacer adjacent motif (PAM) binding assays. It has also been used in the culturing of Agrobacterium tumefaciens and E. coli DH10B.

Components

16 g/L Tryptone
10 g/L Yeast Extract
5 g/L NaCl

Preparation Note

1. Stir to suspend 31 g of 2xYT Microbial Medium Powder in 1 L of water.
2. Heat to boiling while stirring to dissolve all ingredients completely.
3. Autoclave for 15 minutes at 121 °C.

Reconstitution

Add 31 g powder in 1L water. Heat to boiling while stirring to dissolve powder. Autoclave for 15 minutes at 121°C to sterilize. Allow to cool before making additions, such as antibiotics (if desired).
