Gene Information

General description

SDF-1 (stromal-derived factor 1) is a constitutive gene which is ubiquitously expressed in vertebrate tissues. It was first identified in rat bone marrow (BM) stromal cell line supporting hematopoietic cell culture. This gene is regulated at splicing level, rather than transcriptional level and is spliced into multiple variants. Among these, SDF-1α is the major isoform expressed in all tissues, but is rapidly proteolysed in blood. This isoform is composed of 89 amino acids. SDF-1 is a member of the intercrine cytokine family or chemokine family of proteins, which are characterized by four conserved cysteines that form two disulfide bonds.
Recombinant rat SDF-1α is a 7.9kDa protein containing 68 amino acid residues.

Biochem/physiol Actions

SDF-1 (stromal-derived factor 1) performs a key function in cell migration and proliferation and in tissue-specific processes such as neuromodulation. This gene is also involved in chronic inflammatory disorders, metastasis and malignancy, HIV (human immunodeficiency virus) infection and benign proliferative diseases. During ontogeny, SDF-1α is crucial for appropriate patterning, and it promotes growth and differentiation of critical cell types. During pancreatic regeneration, SDF-1α- CXCR4 (chemokine receptor) ligand-receptor pair is crucial for maintaining the duct cell survival, proliferation, and migration.

Physical form

Sterile filtered through a 0.2 micron filter. Lyophilized with no additives.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
