General description

Peroxiredoxin 6 (Prdx6) is a nonseleno peroxidase enzyme, encoded by the gene mapped to human chromosome 1q25.1, a region associated with schizophrenia. Prdx6 belongs to thiol-specific antioxidant protein family. Prdx6 is highly expressed in lung, alveolar epithelial type II cells, bronchiolar Clara cells, and alveolar macrophages. Unlike other members of the family, Prdx6 is characterized with a single conserved cysteine residue and has been called as 1-Cys Prdx. Prdx6 is a ~25,100Da protein containing 224 amino acids.

Biochem/physiol Actions

Peroxiredoxin 6 (Prdx6) is a bifunctional enzyme, carrying out both phospholipase A2 (PLA2) activity and glutathione (GSH) peroxidase function. The encoded protein aids in redox regulation of the cell and is implicated in reduction of H2O2 and short chain organic, fatty acid, and phospholipid hydroperoxides. It might also have a crucial role in the regulation of phospholipid turnover and in antioxidant defense. Prdx6 is highly expressed at leading edges of wounds and aids in wound healing. Deficiency of the enzyme activity is observed in cataracts of the lens. Overexpression of Prdx6 leads to several degenerative CNS (central nervous system) disorders such as Alzheimer′s, Parkinson′s, Pick′s, and Crutzfeld-Jakob diseases and schizophrenia. In addition, elevated expression of the enzyme is also observed in various neoplastic diseases including malignant mesothelioma, bronchogenic squamous cell carcinoma and cancer of the gingivo-buccal area, bladder and breast.

Physical form

1 mg/mL solution in 20 mM Tris-HCl buffer (pH 8.0) containing 20% glycerol.
