General description

HDAC7 (histone deacetylase 7) is located on human chromosome 12q13. It is expressed in fetal and adult pre-B cells.

Application

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.

Biochem/physiol Actions

Histone deacetylases (HDACs) play crucial roles in the epigenetic modulation of gene expression and helps in the growth, differentiation, and apoptosis of cancer cells. Knockdown of HDAC7 stimulates apoptosis and autophagy in salivary mucoepidermoid carcinoma cells. It may play a vital role in both types of B lymphopoiesis. HDAC7 serves as a repressor of myeloid genes.

Unit Definition

One unit is defined as the amount of enzyme required to deacetylate 1 pmol of substrate/min at 37°C.

Physical form

Formulated in 25 mM Tris-HCl, pH 8.0, 138 mM NaCl, 0.05% Tween-20 and 10% glycerol.

Preparation Note

Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.
