Lasmiditan (COL-144 and LY573144) is an orally active, potent and selective high-affinity serotonin receptor 5-HT1F agonist (5-HT1F/1E Ki = 2.21/594 nM; 5-HT1A/B/C Ki >1 μM; 5-HT2A/B/C Ki >2 μM; 5-HT6/7 Ki >3 μM). Lasmiditan induces mitochondrial biogenesis (MB) in human glomerular endothelial cells and mouse glomerular endothelial cells (100 nM for 72h). Unlike the triptans, Lasmiditan is void of vasoconstrictor activity toward isolated human coronary arteries (up to 100 μM) or in anaesthetized female beagle dogs in vivo (4.13-11.13 mg/kg i.v.)
