General description

After the removal of highly abundant proteins (HAP) with the Seppro® mouse depletion column (SEP110 or SEP 090), the next level of protein abundance, the moderately abundant proteins (MAP) remains an obstacle in accessing the (low abundant proteins) LAP, where the majority of biologically interesting and clinically important biomarkers reside. Seppro Supermix Mouse can further partition complex plasma/serum samples. This product is generated by immunizing chickens with mouse plasma proteins from which the 7 HAP have been removed. The resulting antibodies are mainly against MAP and or immunoreactive proteins. Both the collected flow-through fraction and eluted fraction can be used for downstream analysis.

Application

Seppro® Supermix, LC5 (5 mL column) is used as avian polyclonal IgY-based immunoaffinity follow-on separation of mouse serum moderately abundant proteins (MAP) from mouse serum low abundant proteins (LAP) after specific mouse serum highly-abundant proteins (HAP) have been removed using Seppro® mouse depletion column SEP10.

  1. How does the storage temperature relate to shipping conditions?

    The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.   6. How many moderately abundant proteins (MAPs) does Product SEP100, Seppro® Supermix Mouse, deplete?

    As of this time, the exact number of depleted MAPs by SEP100 is not precisely defined.  This question is currently under investigation.

  7. My protein sample contains SDS.  Is this compatible with Seppro® Supermix Mouse?  

    No. We do not recommend using protein samples containing SDS with Product No. SEP100.  SDS is a detergent and would be expected to affect adversely the activity of the conjugated antibody in terms of depletion.
