General description

Monoclonal Anti-Neuron-specific Enolase (NSE) (mouse IgG1 isotype) is derived from the hybridoma NSE-P2 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide. Neuron-specific enolase (NSE) is present in the brain especially in the neuroendocrine cells and neurons. It is a cytosolic enzyme, that exists as a dimer and belongs to the family of enolase enzymes. NSE gene is mapped to human chromosome12p13.31.

Specificity

Monoclonal Anti-Neuron-specific Enolase (NSE) recognizes human and rat NSE.

Immunogen

synthetic peptide corresponding to an internal sequence of human NSE.1 The isotype is determined by ELISA using Mouse Monoclonal Antibody Isotyping Reagents (Sigma ISO-2).

Application

Monoclonal Anti-Neuron-Specific Enolase (NSE) antibody produced in mouse may be used in:
  • immunoblotting
  • enzyme-linked immunosorbent assay (ELISA)
  • immunohistochemistry

Biochem/physiol Actions

Neuron-specific enolase (NSE) catalyzes the production of phosphoenolpyruvate from 2-phospho-D-glycerate in the glycolytic pathway. NSE is a specific neurobiochemical marker of brain damage after traumatic brain injury, hypoxic encephalopathy and stroke. Moreover, NSE has been suggested to be a general circulating marker for neuroendocrine tumors. Interestingly, high levels of NSE were also reported in breast cancer upon exposure to the environmental pollutants arsenite and cadmium.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For extended storage, freeze at -20 °C in working aliquots. Repeated freezing and thawing or storage in “frost-free” freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
