General description

Monoclonal Anti-ADFP/PLIN2 (mouse IgG1 isotype) is derived from the hybridoma ADFP-5 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide.
Perilipin 2 (PLIN2) is a 50 kDa major protein belonging to perilipin family. It is present on lipid droplets found in the cytoplasm of variety of cells as hepatocytes, adipocytes, muscle cells, mammary epithelial cells, fibroblasts, endothelial cells and macrophages. PLIN2 gene, spanning 14 kb, with eight exons, is located on human chromosome 9p22.1.

Specificity

Monoclonal Anti-ADFP/PLIN2 recognizes human ADFP/PLIN2.

Immunogen

a synthetic peptide corresponding to a sequence at the C-terminus of human PLIN2, conjugated to KLH.

Application

Monoclonal Anti-ADFP / PLIN2 antibody produced in mouse may be used in immunoblotting and immunofluorescence.

Biochem/physiol Actions

PLIN2 may serve as a biomarker of lipid accumulation in diverse cell types and diseases.
Perilipin 2 (PLIN2) plays a vital role in uptake and storage of lipids as triacylglycerols in lipid droplets in skeletal muscle. PLIN2 is also involved in the regulation of lipid metabolism. High levels of PLIN2 is beneficial for the prevention of insulin resistance. Overexpression of PLIN2 is associated with clear cell renal cell carcinoma (CC-RCC).

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8°C for up to one month. For extended storage, freeze at -20oC in working aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
