General description

IQ motif containing GTPase activating protein 1(IQGAP1) is encoded by the gene mapped to human chromosome 15q26.1. The encoded 190kDa protein is widely expressed and is mainly localized to lamellipodia and membrane ruffles in fibroblasts and other cell types.
IQGAP1 is a scaffolding protein and is made up of N-terminal calponin-homology domain (CHD), WW domain and guanosine triphosphatase (GTPase) activating protein GAP-related domain (GRD).

Specificity

Anti-IQGAP1 (C-terminal region) specifically recognizes human IQGAP1.

Application

Anti-IQGAP1 (C-terminal region) antibody produced in rabbit may be used in:

  • western blotting
  • immunohistochemistry

Biochem/physiol Actions

IQ motif containing GTPase activating protein 1(IQGAP1) interacts with cell division control protein 42 homolog (Cdc42) or Rac1 and regulate cadherin-based cell adhesion. IQGAP1 binds to NLRC3 in human monocytic and epithelial cells and regulate type I interferon (IFN) production. Overexpression of the gene has been observed in gastric cancer cell lines.In addition, IQGAP1 is also associated with diseases, such as cancer and diabetes. The encoded protein is also implicated in the production and secretion of insulin. IQGAP1 also aids in the endocytosis and plasma membrane fusion of vesicles in pancreatic β-cells.
IQGAP1 plays a role in tumorigenesis. IQGAP1 gene is upregulated in several carcinomas, including colorectal, lung, ovary, and breast carcinoma.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

Store at –20 °C. For continuous use, store at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
