General description

Monoclonal Anti-DNA polymerase δ p50 (rat IgG2a isotype) is derived from the hybridoma PDK 7B4 produced by the fusion of mouse myeloma cells (P3X63Ag8.653) and splenocytes from rat immunized with human p50. DNA polymerase δ consists of the proliferating cell nuclear antigen (PCNA), the multi-subunit replication factor C, and the 4 subunit DNA polymerase complex: DNA Polymerase δ 1, 2, 3, 4 (POLD1, POLD2, POLD3 and POLD4). The core DNA polymerase δ exists as a heterodimeric enzyme with subunits of 125 and 50 kDa.
The DNA polymerase δ 2 subunit (POLD2) gene, with 11 exons spanning 10kb of genomic DNA, is mapped to human chromosome 7. The gene encoding 50kDa protein is characterized with the G+C-rich 5′-flanking region and it lacks a typical TATA box. The encoded protein is one of the components of human DNA Pol δ complex.

Biochem/physiol Actions

DNA polymerase δ 2 subunit (POLD2) gene codes for a protein that participates in DNA replication and repair. In COS-7 cells, regulatory region of the hPOLD2 gene has ability to facilitate the expression of chloramphenicol acetyltransferase reporter gene. Phosphatase and tensin homolog (PTEN) tumor suppressor gene decrease the expression of POLD2 gene associated with ovarian carcinogenesis. This protein might also function as a potential prognostic biomarker in ovarian carcinoma. POLD2 takes part in polymeraseδ (Pol δ) assembly by coordinating simultaneously with all other three subunits of the enzyme.
