General description

SYPRO Tangerine Protein Gel Stain is a versatile stain for proteins in SDS gels. The staining procedure is fast, simple and sensitive and does not require the use of acids or organic solvents; the staining can be done in saline solution. Proteins stained without fixation can be used for in-gel enzyme activity assays if SDS does not inactivate the protein of interest. Stained proteins can also be eluted from gels for further analysis. Since the stain does not alter protein structure, it does not interfere with analysis by mass spectrometry. Nor does it interfere with the transfer of proteins to blotting membranes, thus allowing visualization of proteins prior to blotting. The dye is compatible with fixation using 7% acetic acid or 12.5% TCA.
SYPRO® Tangerine Protein Gel Stain is:
  • Highly sensitive. The stain can detect 4 to 8 ng of protein per minigel band, making it more sensitive than Coomassie® Brilliant Blue staining and as sensitive as many silver staining techniques.
  • Rapid. Staining is complete in less than one hour.
  • Simple. After electrophoresis, the gel is stained, rinsed and photographed; no fixation or destaining steps are required.
  • Compatible with standard laboratory equipment. Stained proteins can be visualized with a standard 300 nm UV transilluminator or laser scanner.
  • Cost-effective. Staining with SYPRO Tangerine is less expensive than silver staining and requires less time.
  • Low protein-to protein variability. The dye interacts with the SDS coat around the protein, giving more consistent staining between different types of proteins compared to Coomassie or silver staining.
  • Selective for proteins. SYPRO Tangerine detects proteins as small as 6.5 kDa and does not stain nucleic acids or lipopolysaccharides.
  • Broad linear range of detection. The stain interacts with the SDS-protein micelle during gel staining at a fairly constant stoichiometry of 1.4 to 1, such that the fluorescence intensity of the stained bands is linear with protein quantity over three orders of magnitude (a much broader range than either Coomassie or silver staining).
