General description

Granzyme B (GZMB; EC 3.4.21.79) is a serine protease stored in the secretory granules of leukocytes. It induces cytotoxic T lymphocyte (CTL)-mediated apoptosis by the activation of caspases and the degradation of structural proteins of the target cells. Granzyme B has an important role in CTL-mediated immune responses and has been implicated in chronic inflammatory diseases such as rheumatoid arthritis, coronary syndrome, and chronic angina pectoris.

Principle

The Granzyme B Inhibitor Screening Kit is a simple and high-throughput assay for screening potential inhibitors of granzyme B. Granzyme B activity is measured by cleaving a synthetic 7-amino-4-trifluoromethylcoumarin (AFC)-based substrate to yield AFC, a fluorescent product (λex = 380/ λem = 500 nm), proportional to the enzymatic activity present. The substrate includes the recognition sequence for granzyme B.
