MTT Formazan has been used to measure cell viability.

Biochem/physiol Actions

MTT assay is a colorimetric reaction, generally used to examine cell viability. It involves the enzymatic reduction of a yellow-colored, water-soluble, 3-[4,5-dimethylthiazole-2-yl]-2,5-diphenyltetrazolium bromide (MTT) to purple-colored, water-insoluble, MTT-formazan. This reaction is catalyzed by mitochondrial dehydrogenase and only occurs in viable cells. Thus, the amount of blue formazan is proportional to the number of viable cells. The absorbance of the formazan solution is analyzed spectrophotometrically at 570 nm.

    No.  MTT or thiazolyl blue tetrazolium is used to determine the number of viable cells in well-cultures. Consider use of kit CGD1 or product M5655.

  4. What is the solubility of MTT Formazan, Product M2003, in water or PBS?

    This product is not soluble in either water or PBS.  Sigma Aldrich tests product at 1mg/mL in acetone. A 1:1 DMSO:acidified isopropanol also works well for dissolving the formazan.

  5. Can MTT Formazan, Product M2003, be used to measure dehydrogenases?

    No. MTT is converted by active mitochondrial dehydrogenases of living cells to MTT Formazan. Consider use of product M2128 or M5655.

  6. What is the proper wavelength for measuring MTT Formazan, Product M2003?

    We recommend detection at 570 nm, with background subtraction at 630-690 nm in acidified isopropanol.
