General description

Mucolipin-2 (MLN2) is a member of transient receptor potential (TRP) protein superfamily. It consists of gated, tetrameric cation channels. MLN2 has a conserved structure of six transmembrane helices with differing cytoplasmic oriented N- and C-terminal domains. It is localized to late endosomes and lysosomes..

Specificity

Additional bands may be observed at ~120 kDa and ~35 kDa due to aggregation and degradation, respectively.

Immunogen

synthetic peptide corresponding to amino acids 534-549 of human mucolipin-2. This sequence has low homology with rat (73% identity) and mouse (66% homology) mucolipin-2.

Application

Anti-Mucolipin-2 (C-terminal) antibody produced in rabbit has been used in immunoblotting.

Biochem/physiol Actions

Mucolipin-2 (MLN2) plays an important role in lysosome maturation, lysosome fission, late endosome-lysosome fusion, autophagosome-lysosome fusion, lysosome expulsion and iron release. It stimulates autophagy proliferation via transcription factor EB (TFEB) activation. MLN2 is involved in B‐lymphocyte development.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
