General description

Tripartite motif 65 (TRIM65) belongs to TRIM family proteins. TRIM family contains B-box 1 and a B-box 2 type domain. The coiled domain possesses hyper-helical regions. TRIM65 exhibits a SPRY (spla and ryanodine) domain at its C-terminus. Two anti-parallel β-sheets in the SPRY domain form a curved β-sandwich. Trim65 homologous genes have been found in mammals.

Biochem/physiol Actions

Ubiquitination of TNRC6 proteins is through E3 ligase activity of TRIM65. The coiled coil domain plays a role in protein-protein interactions and oligomerization. It is associated with repression of miRNA activity. TRIM65 colludes and adjoins with trinucleotide repeat TNRC6 (trinucleotide repeat-containing gene 6) proteins. Genomic studies on cerebral white matter lesions have found a new locus on chromosome 17q25 accommodating TRIM65. The studies also determine a probable mechanism leading to these lesions. Uncontrolled TRIM65 activities extirpate TNRC6 stability and alter RISC (RNA-induced silencing complex) stability, miRNA activity and cellular homeostasis. Gene silencing of TRIM65 with that of siRNA leads to attenuation of let-7a reporter activity. An abnormal expression of TRIM65 has a similar effect.
