General description

BEN domain-containing protein 3 (BEND3) is localized in the nucleoli of cells. This heterochromatin-associated protein possesses three amino acid residues at the amino-terminal region which functions as a nuclear localization signal (NLS).

Application

Biochem/physiol Actions

BEN domain-containing protein 3 (BEND3) plays a role as a transcriptional repressor in the nucleus wherein it binds to ribosomal RNA (rRNA) gene promoters to aid in suppressing rRNA genes. Loss of function of BEND3 has shown a decrease in the methylation of rDNA promoters. The protein associates with the nucleolar-remodeling complex (NoRC) and in its SUMOylated form, it associates with ubiquitin specific protease 21 (USP21) debiquitinase. Its overexpression causes early chromatin condensation and vigorous heterochromatinization, leading to cell cycle arrest.
