General description

TPBG (trophoblast glycoprotein) is an oncofetal glycoprotein with a molecular weight of 72kDa. It is rarely expressed in normal adult tissues, but it is seen to be expressed in multiple cancerous tissues. It is also expressed by trophoblast cells and placenta. It is a cell surface protein. Its extracellular domain is highly glycosylated and contains eight leucine-rich repeats (LRRs). This protein is composed of 420 amino acids and contains a single transmembrane domain.

Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. Biochem/physiol Actions

TPBG (trophoblast glycoprotein) interacts with human trophoblast. This protein is highly expressed in pancreatic ductal adenocarcinoma (PDAC), which is linked with tumor-node-metastasis (TNM) stage of the tumor. It is also involved in metastasis in PDAC. This protein promotes EMT (epithelial-mesenchymal transition) and CXCL12/CXCR4 (C-X-C motif chemokine ligand/CXC receptor) chemotaxis, and inhibits canonical Wnt/β-catenin. It is overexpressed in malignant pleural mesothelioma (MPM), and has potential as an immunotherapy target in the same.
