General description

ALKBH3 (alkB homolog 3, α-ketoglutarate-dependent dioxygenase), also known as prostate cancer antigen (PCA)1, shows high homology to E. coli alkB protein. It is a DNA and RNA repair enzyme.

Application

Biochem/physiol Actions

ALKBH3 (alkB homolog 3, α-ketoglutarate-dependent dioxygenase) is overexpressed in non-small-cell lung cancer and results in increased cell proliferation. Inactivation of this protein leads to the activation of p21 and p27, which causes cell cycle arrest. It interacts with NOX (nitric oxide)-2-ROS (reactive oxygen species) and Tweak (TNF homologue with weak apoptosis-inducing activity)/Fn14 (fibroblast growth factor-inducible protein 14)-VEGF (vascular endothelial growth factor) signaling pathways and promotes survival, angiogenesis, and invasion in urothelial carcinomas. It has potential as a diagnostic marker for prostate cancer (PC), and might be one of the more aggressive variants of PC. It promotes tumorigenesis in pancreatic cancer by facilitating angiogenesis and resistance to apoptosis.
