General description

PNKP (Polynucleotide kinase 3′-phosphatase) is a 57.1kDa, DNA repair protein. It consists of ATP binding, kinase domain at the C-terminal end, a potential 3′-phosphatase central domain and a fork-head associated (FHA) domain at the N-terminus end.

Application

Biochem/physiol Actions

PNKP (Polynucleotide kinase 3′-phosphatase) is majorly involved in the DNA repair caused by ionizing radiation or oxidative damage. It possesses two enzymatic activities, a DNA 5′ kinase activity and a 3′ phosphatase activity. In DNA repair, it functions in the two processes, single-strand break repair and non-homologous telomeric end joining for double-strand break repair. Mutation in the PNKP gene mainly alters the DNA repair process and also causes microcephaly and seizures.
