General description

CD4, a glycoprotein, is predominantly expressed on the cell membrane of mature, thymus-derived (T) lymphocyte and also to some extent in the monocyte/macrophage lineage. It has a transmembrane domain, cytoplasmic region and an extracellular region of four tandem domain similar to members of Ig (immunoglobin) superfamily.

Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. These images and the collection of this vast data set can be viewed on the Human Protein Atlas (HPA) site by clicking on the Image Gallery link. Biochem/physiol Actions

CD4 (cluster of differentiation 4) interacts with the CD4+ T cells with antigen-presenting cells or with target cells by binding to the major histocompatibility complex (MHC) class II molecules. Interaction with MHC-II molecules increases the cytokine expression and the differentiation of human monocytes into functional mature macrophages. Further, it effects human immunodeficiency virus (HIV) binding and subsequent infection. Thus, it proves an important role in HIV as the primary receptor.
