Application

Hyaluronic acid sodium salt from rooster comb has been used:
  • as a component of bi-layer gelatin scaffold
  • as a vehicle for release of gentamicin during left ear surgery in guinea pigs
  • in hyaluronic acid binding assay in human embryonic kidney cell line 293T

Biochem/physiol Actions

High molecular mass polymer composed of repeating dimeric units of glucuronic acid and N-acetyl glucosamine which forms the core of complex proteoglycan aggregates found in extracellular matrix.
Hyaluronic acid is a polysaccharide present in the skin, connective tissue, vitreous humor and synovial fluid. It is essential for tissue hydration and functions to regenerate skin post injury or wounds. Hyaluronic acid, apart from having dermatologic and ophthalmologic applications, serves as a drug delivery vehicle. Hyaluronic acid also exhibits antioxidant and antiglycation properties.

    Product No. H5388 is a linear heteropolysaccharide with repeating 3-O-(β-D-glucuronido)-N-acetyl-D-glucosamine units linked by β-(1-->4)) hexosaminidic bonds.  The molecular weight of this subunit has not been determined, but could be calculated from the information above. Molecular weights of hyaluronic acids vary from 0.1-10 million, depending on the source.  The hyaluronic acid from rooster comb is expected to have a molecular weight of 1-4 million.

  6. What is the purity of Hyaluronic acid, Product H5388?

    Sigma does not perform purity testing on this product due to the variable molecular weight and degradation that may occur in native glycosaminoglycans.  This product is tested for activity and protein content which provide information regarding impurities and suitability.

  7. What is the solubility of Hyaluronic acid, Product H5388?

    For Sigma's analytical test, 15 mg of this product is dissolved in 5 mL of 0.3 M phosphate buffer, pH 5.35 at 25°C (i.e., 3 mg/mL).

Protocols

To measure hyaluronidase activity, a turbidimetric determination assay is used at 600 nm. One unit of hyaluronidase activity will cause a change in absorbance of 0.330 per minute at pH 5.35 at 37 °C.
