Biochem/physiol Actions

The lac operon (lactose operon) is an essential operon used for the transport and metabolism of lactose in Escherichia coli and some enteric bacteria. It has three adjacent structural genes, lacZ, lacY and lacA. The lacZ gene encodes β-galactosidase. Escherichia coli β-galactosidase is mostly used as a reporter enzyme. It is produced under oxidizing conditions in the cytoplasm of an Origami strain. β-galactosidase occurs in a disulfide bond. At the active site of β-galactosidase a loop opens and closes during catalysis.

Ghrelin plays a key role in appetite, energy homeostasis, and glucose regulation. Recent evidence suggests ghrelin suppresses inflammation in obesity; however, whether this is modulated by the acylated and/or des-acylated peptide is unclear. We used mice deficient in acylated ghrelin

Escherichia coli β-galactosidase is probably the most widely used reporter enzyme in molecular biology, cell biology, and biotechnology because of the easy detection of its activity. Its large size and tetrameric structure make this bacterial protein an interesting model for
