Application
Iodoacetamide has been used:
- as a component in urea-sodium dodecyl sulfate (SDS) buffer for yeast cell lysis in immunoprecipitation assay
- in equilibration of pH gradient strips during the second dimension run in isoelectric focusing
- as a protein alkylation agent
Biochem/physiol Actions
Iodoacetamide (IAA) modifies thiol groups in proteins by S-carboxyamidomethylation. Iodoacetamide is an alkylating agent. IAA is commonly used to analyze the key role of cysteine residues in the catalytic reactions of enzymes. It is also used in the derivatization of cysteine residues in proteins for proteomic analysis. IAA irreversibly inhibits glyceraldehyde-3-phosphate dehydrogenase (GAPDH).
