General description
Glucose-6-phosphatase is a multicomponent protein complex comprising of glucose-6-phosphate transporter (G6PT) and a catalytic unit (G6PC). Liver and kidney cells show predominant expression of glucose-6-phosphatase.
Application
Glucose-6-Phosphatase from rabbit liver has been used in glucose 6-phosphatase inhibitory assay of crude C. anisata leaf extracts and methanolic leaf and bark extracts of Dolichandrone atrovirens.
Glucose-6-Phosphatase was used to study the effects of Clausena anisata (Wild hook leaf) extracts on selected diabetes-related metabolizing enzymes.
Biochem/physiol Actions
Glucose-6-Phosphatase is crucial for regulating glucose homeostasis during starvation. Deficiency of glucose-6-phosphatase is implicated in glycogen storage disease type I (GSDI).
Glucose-6-phosphatase catalyzes the conversion of glucose-6-phosphate into glucose as the last step of gluconeogenesis.

It has been reported that glucose-6-phosphatase, inorganic pyrophosphatase, and pyrophosphate glucose phosphotransferase are associated with a single protein that catalyzes three different reactions:
a) glucose 6-phosphate + H2O → glucose + Pi
b) PPi + H2O → 2 Pi
c) glucose + PPi or nucleoside-DP or -TP → glucose 6-phosphate + Pi or nucleoside-MP.
a) glucose 6-phosphate + H2O → glucose + Pi
b) PPi + H2O → 2 Pi
c) glucose + PPi or nucleoside-DP or -TP → glucose 6-phosphate + Pi or nucleoside-MP.
One unit will release 1.0 μmole of inorganic phosphate from glucose 6-phosphate per min at pH 6.5 at 37 °C.
