General description

Dulbecco′s Modified Eagle′s medium (DMEM), a regularly used cell culture medium, is a source of energy-rich media that includes glucose, vitamins, and amino acids. A 1:1 mixture of Dulbecco′s Modified Eagle′s medium (DME) and Ham′s F-12 nutrient mixture is commonly used in cell culture research. To compensate for the loss of buffering capacity caused by removing serum, 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (HEPES) buffer may be added in the formulations at a final concentration of 15 mM.

    This is a 1:1 mixture of the DME and F-12. The optimal carbon dioxide required for DME is 10% and for F-12 is 5%. Since this medium is a mixture, the optimal carbon dioxide concentration is typically 5% to 8%.

  5. Can Product D8437, Dulbecco’s Modified Eagle’s Medium/Nutrient Mixture F-12 Ham, be used under serum free conditions?

    When DME/F12 is used under serum free conditions, supplementation is necessary. DME/F12 can be used for growing the epithelial cell line TM4 (mouse testes) when supplemented with insulin, transferrin, EGF, FSH, somatomedin C, ovine growth hormone, and retinoic acid. Primary cultures of Sertoli cells can also be grown in the above supplemented media. Rat glial C6 cells have been grown in DME/F12 supplemented with transferrin, insulin, LH, and rat submaxillary gland extract.
