General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Evaluated by Flow Cytometry in LPS-stimulated mouse macrophages.

Flow Cytometry Analysis: One million BALB/c mouse macrophages were stimulated with 1 μg/mL of LPS for 24 hours and stained with 5 μL of a 1:1 mixture of Cat. No. CWA-1155, Anti-Mouse TIM-4 (RMT4-54) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of Cat. No. CWA-1018 Rat IgG2a isotype control and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye.

Tested Applications

Flow Cytometry Analysis: One million BALB/c mouse macrophages were unstimulated (left figure) or stimulated (right figure) with 1 μg/mL of LPS for 24 hours and stained with 5 μL of a 1:1 mixture of Cat. No. CWA-1155, Anti-Mouse TIM-4 (RMT4-54) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-APC ColorWheel® Antibody-Ready Allophycocyanin (APC) Dye or an equivalent amount of Cat. No. CWA-1018 Rat IgG2a isotype control and Cat. No. CWDS-APC ColorWheel® Antibody-Ready Allophycocyanin (APC) Dye.

Target description

T-cell immunoglobulin and mucin domain-containing protein 4 (UniProt: Q6U7R4; also known as TIMD-4, Spleen mucin-containing knockout of lymphotoxin protein, SMUCKLER, T-cell immunoglobulin mucin receptor 4, TIM-4, T-cell membrane protein 4) is encoded by the Timd4 (also known as Tim4) gene (Gene ID: 276891) in murine species. TIM-4 is a single-pass type I, homodimeric membrane protein that is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 23-279), a transmembrane domain (aa 280-300), and a cytoplasmic domain (aa 301-343). It also contains an Ig-like type V domain (aa 23-128). It is predominantly expressed in lymphoid tissues, such as spleen, lymph nodes, and in Peyer patches. Its expression is also observed in fetal liver, salivary gland, and spleen stromal cells. TIM-4 serves as a phosphatidylserine (PS) receptor and mediates phagocytosis of apoptotic cells by binding to PS exposed on the surface of these cells. TIM-4 is shown to form a trimeric complex with fibronectin 1 and integrins, which enables efficient recognition of apoptotic cells and mediates their ingestion through the action of integrins. It also acts as a scavenger receptor and mediates phagocytosis of exogenous particles in a phosphatidylserine-independent manner. (Ref.: Min, C., et al. (2020). Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
