General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of Cat. No. CWA-1012 Rat IgG2b isotype control and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye.

Tested Applications

Target description

T-cell surface glycoprotein CD3 gamma chain (UniProt: ; also known as T-cell receptor T3 gamma chain, CD3g) is encoded by the CD3g gene (Gene ID: 12502) in murine species. CD3g is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 23-116), a transmembrane domain (aa 117-137), and a cytoplasmic domain (aa 138-182). When antigen presenting cells (APCs) activate T-cell receptor (TCR), TCR-mediated signals are transmitted across the cell membrane by the CD3 chains CD3D, CD3E, CD3G and CD3Z. All CD3 chains contain immunoreceptor tyrosine-based activation motifs (ITAMs) in their cytoplasmic domain. The ITAM domain of CD3g is localized to amino acids 149-177. Upon TCR engagement, these motifs are phosphorylated by LCK and FYN resulting in the activation of downstream signaling pathways. The absence of CD3g is reported to result in severe reduction in the level of the TCR-CD3 at the cell surface of thymocytes and peripheral T cells. The development of both the TCR β and TCR lineages are affected by the absence of CD3g. Mutations in CD3g gene have been linked to immunodeficiency 17, an autosomal recessive disorder characterized by autoimmune disease and partial T-cell lymphopenia.

Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
