General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Specificity

Clone 5H10-27 is a rat monoclonal antibody that detects mouse Integrin 5 (CD49e). It targets an epitope within the extracellular domain.

Evaluated by Flow Cytometry in C57Bl/6 bone marrow cells .

Staining of one million C57BL/6 mouse bone marrow cells was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1139, Anti-Mouse Integrin 5/CD49e (5H10-27) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-APC ColorWheel® Antibody-Ready Allophycocyanin (APC) Dye or a 1:1 mixture of an equivalent amount of Cat. No. CWA-1018 Rat IgG2a isotype control and Cat. No. CWDS-APC ColorWheel® Antibody-Ready Allophycocyanin (APC) Dye.

Tested Application

Flow Cytometry Analysis: Staining of one million C57BL/6 mouse bone marrow cells was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1139, Anti-Mouse Integrin 5/CD49e (5H10-27) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of Cat. No. CWA-1018 Rat IgG2a isotype control and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye.

Target description

Integrin alpha-5 (UniProt: P11688; also known as CD49 antigen-like family member E, Fibronectin receptor subunit alpha, Integrin alpha-F, VLA-5, CD49e) is encoded by the Itga5 gene (Gene ID: 16402) in murine species. Integrins are heterodimeric integral membrane proteins composed of an -subunit and a β-subunit that function in cell surface adhesion and signaling. Integrin 5/CD49e is expressed on thymocytes, T cells, monocytes, platelets, early B cells, and activated in B cells. It exhibits an increased expression in mesodermal progenitors and is shown to be prevalent in fetal cardiomyocytes. Its deletion is shown to cause mesodermal defects during embryogenesis in murine models. Membrane fractions obtained from fibroblasts of diabetic animals display significant reduction in integrin 5/CD49e levels. Integrin 5/CD49e is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-44), which is subsequently cleaved off to generate the mature form that contains a heavy chain (aa 45-898) and a light chain (aa 899-1053). Structurally, it consists of an extracellular domain (aa 45-999), a transmembrane domain (aa 1000-1025), and a short cytoplasmic domain (aa 1026-1053). It contains four calcium binding sites (aa 480-488; 334-342; 401-409; and 465-473). When associated with β1 subunit it can serve as a receptor for fibronectin and fibrinogen. It recognizes the sequence R-G-D in its ligands. Integrin 5/β1 is reported to bind to PLA2G2A via a site (site 2) that is distinct from the classical ligand-binding site (site 1) and this induces integrin conformational changes and enhanced ligand binding to site 1. Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
