General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Specificity

Clone RB6-8C5 is a rat monoclonal antibody that recognizes a common epitope on LY-6G and Ly-6C.

Quality Control Testing:

Evaluated by Flow Cytometry in Mouse bone marrow cells.

Flow Cytometry Analysis: Staining of one million mouse bone marrow cells was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1051, Anti-Mouse Ly-6G (RB6-8C5) ColorWheel® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Rat IgG2b isotype control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

Lymphocyte antigen 6G/6C (UniProt: P35461/P0CW02; also known as Ly-6G/Ly-6C, Ly-6G.1/Ly-6C.1, Gr-1) are encoded by the Ly6g/Ly6c1 gene (Gene ID: 556644/17067) in murine species. Ly-6 molecules are encoded by multiple tightly linked genes located on mouse chromosome 15 and have conserved genomic organization. They are expressed on fully differentiated cells in specific compartments of peripheral lymphoid tissues. Ly-6 proteins are widely used as differentiation markers on hematopoietic cells. Ly-6G and LY-6C are GPI-anchor proteins that are synthesized with a signal peptide (aa 1-26) and a propeptide (aa 106-134 for LY-6G and 110-131 for LY-6C) that are subsequently cleaved off to produce the mature form. They contain one UPAR/Ly6 domain that is localized in amino acids 27-118 in Ly-6G and in amino acids 27-115 in LY-6C. Ly‐6G is expressed on granulocytes, a subset of eosinophils, and transiently during developmental stages of monocytes. Its expression in bone marrow is directly correlated with granulocyte differentiation and maturation. Clone RB6-8C5 recognizes a common epitope on Ly-6G and LY-6C. In murine cells, this clone can be used in combination with a macrophage labeling antibody, such as anti-CD11b, for phenotypic analysis of monocytes, macrophages and granulocytes. Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
