General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Specificity

Clone RVS-10 is a mouse monoclonal antibody that detects human Transferrin receptor protein 1 (CD71).

Immunogen

Full-length human Transferrin receptor protein 1 (CD71).

Application

Quality Control Testing:

Evaluated by Flow Cytometry in K562 cells.

Flow Cytometry Analysis: Staining of one million human K562 cells was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1039, Anti-Human CD71 (RVS-10) ColorWheel® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel® activated Phycoerythrin (PE) Dye or an equivalent amount of Mouse IgG1 Isotype Control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

Transferrin receptor protein 1 (UniProt: P02786; also known as TR, TfR, TfR1, Trfr, T9, p90, CD71) is encoded by the TFRC gene (Gene ID: 7037) in human. Iron is essential for multiple metabolic processes and is incorporated into many proteins in the form of cofactors such as heme and iron-sulfur clusters. CD71 (Transferrin receptor 1) facilitates the uptake of iron at the cell surface by internalizing diferric transferrin. It is a single-pass type II membrane protein that contains a cytoplasmic domain (aa 1-67), a transmembrane domain (aa 68-88), and a long extracellular domain (aa 89-760). Cellular uptake of iron occurs via receptor-mediated endocytosis of ligand-occupied transferrin receptor into specialized endosomes where acidification leads to iron release. The apotransferrin-receptor complex is then recycled to the cell surface with a return to neutral pH and the concomitant loss of affinity of apotransferrin for its receptor. The transferrin/CD71 system is shown to be important for erythroblasts and lymphocytes, T and B cells and its deficiency leads to anemia and compromised immune responses. Mutations in TFRC gene have been linked to immunodeficiency 46 that is characterized by early-onset chronic diarrhea, recurrent infections, intermittent neutropenia, and thrombocytopenia. Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
