General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Specificity

Clone HI100 is a mouse monoclonal antibody that detects human CD45RA.

Immunogen

Human Receptor-type tyrosine-protein phosphatase C (CD45RA).

Application

Quality Control Testing:

Evaluated by Flow Cytometry in Human peripheral blood mononuclear cells (PBMC).

Flow Cytometry Analysis (FC): Staining of one million Human peripheral blood mononuclear cells (PBMC) was performed using 5 μL of 1:1 mixture of Cat. No. CWA-1000, Anti-Human CD45RA (HI100) ColorWheel® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of a PE-conjugated Mouse IgG2b isotype control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

Receptor-type tyrosine-protein phosphatase C (UniProt: P08575; also known as EC:3.1.3.48, Leukocyte common antigen, L-CA, T200, CD45) is encoded by the PTPRC (also known as CD45) gene (Gene ID: 5788 in human. CD45 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 26-577), a transmembrane domain (aa 578-598), and a cytoplasmic domain (aa 599-1306). It also contains two tyrosine phosphatase (PTPase) domains that are localized to amino acids 653-912 and 944-1228. The first PTPase domain has enzymatic activity, while the second PTPase domain interacts with and stabilizes the first domain and can affect its substrate specificity. CD45 is shown to dephosphorylate Fyn and Lck at their C-terminal tyrosine residues and upregulate their kinase activity. It can also inhibit Fyn and Lck activities by dephosphorylating their regulatory autophosphorylation site. CD45 expression is observed in lymphocytes, eosinophils, monocytes, basophils, and neutrophils. Several isoforms of CD45 have been described that are generated through alternative splicing. It is shown to be essential for T-cell activation through the antigen receptor. It acts as a positive regulator of T-cell co-activation upon binding to DPP4. Isoform CD45RA is expressed on approximately 50% of all peripheral CD4+ and CD8+ T-cells and on a some B cells and monocytes. It functions to enhance signaling through both the T-cell receptor and B-cell receptor. Clone HI100 is widely used as a leukocyte marker for naïve and activated T-cells where naive T cells are positive for CD45RA and activated T-cells are negative for CD45RA.

Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
