General description

Chondroitin sulfate is a linear polysaccharide composed of alternating disaccharide glucuronic acid and N-acetylgalactosamine. It contains proteoglycan (PG) of the extracellular matrix (ECM). Monoclonal Anti-Chondroitin Sulfate (mouse IgM isotype) is derived from the hybridoma produced by the fusion of mouse myeloma cells and splenocytes from an immunized mouse.
This product can be found as purified product that was produced using cell culture hybridoma (SAB4200696)

Specificity

The antibody has been reported to be specific for the glycosaminoglycan (GAG) portion of native chondroitin sulfate proteoglycan (CSPG). The antibody reacts specifically with chondroitin sulfate types A and C but not with type B (dermatan sulfate), and may be used for localization of chondroitin sulfate in tissues and cultured fibroblasts.

Application

Monoclonal Anti-Chondroitin Sulfate antibody produced in mouse was used:
  • in immunohistochemistry as primary antibody to study of spinal cords and brain cryosections.
  • in ELISA and western blot for purification of soluble forms of myelin-associated glycoprotein (MAG) and neural cell adhesion molecule (N-CAM).
  • in flow cytometry studies.
  • as primary antisera in a study of TGF-β regulation.
Monoclonal Anti-Chondroitin Sulfate can also be used to study chondroitin sulfate proteoglycan (CSPG) distribution and its relationships to specific cell-substrate contacts. It is also suitable for microarray and indirect immunofluorescence at the dilution of 1:200 by using bovine mammary gland epithelial (BMGE) cells.

Biochem/physiol Actions

Chondroitin sulfate is involved in tumor invasion and also plays a vital role in stabilizing cell-substratum interactions during early events of melanoma cell spreading on endothelial basement membranes. In Chondroitin sulfate proteoglycan (CSPG), proteoglycan part is involved in cell proliferation and migration which stimulates endothelial cells motility during microvascular morphogenesis. Chondroitin sulfate is used in osteoarthritis treatment. Upregulation of chondroitin sulfate proteoglycans is observed after neuronal injury. It possess the ability to inhibit axonal regeneration.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month.
For extended storage, the solution may be frozen in working aliquots. Repeated freezing and thawing is not recommended. Storage in "frost-free" freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.
