  5. What molecular weight range is CM Sephadex® C-50, Product C50120, suitable for?

Sephadex C-50 is suitable for larger biomolecules such as proteins in the molecular weight range of 30 to 100 kDa.

  6. How do I prepare CM Sephadex® C-50, Product C50120?

Weigh out the required amount of dry powder and suspend it in binding buffer. In general, 1 gram of dry powder CM Sephadex C-50 will give about 30 mL final volume, when swollen in saline buffer.  Swelling should be done at the pH at which the resin is going to be used. Complete swelling takes 1-2 days at room temperature, or 2 hours at 100°C over boiling water.

  7. How do I regenerate CM Sephadex® C-50, Product C50120?

Depending on the nature of the sample, regeneration is normally preformed by washing with a high ionic strength buffer (1-2 M NaCl) and/or increasing pH, followed by re-equilibrating in binding buffer.
