Will CelLytic™ M, Product No. C2978, interfere with protein concentration assays such as Bradford, Lowry and BCA?
We have tested and found CelLytic™ M to be compatible for Bradford Assay and BCA assays. For the Lowry method, there is a background issue that is problematic for low protein concentrations, but may be subtracted with high protein concentrations.
Will Product No. C2978, CelLytic™ M solubilize membrane proteins?
CelLytic™ M can solubilize membrane proteins as well as cytoplasmic proteins and the conditions are non-denaturing. However, we cannot guarantee the solubilization of all membrane proteins. The following proteins were detected in the CelLytic™ M lysate when tested with these antibodies against membrane proteins: Placental alkaline phosphatase, CD45, Connexin and Caveolin.
Is there any denaturants in Product C2978, CelLytic™ M that can interfere with my downstream applications?
CelLytic™ M does not contain any primary amines; it is a buffered solution (pH 7.6) which has no denaturants except for a mild detergent. The exact formulation is proprietary. This product may be diluted or dialyzed if it is found to have interference in your particular assay.
Does Product C2978, CelLytic™ M contain any protease inhibitors?
No, CelLytic™ M does not contain protease inhibitors. We recommend the addition of one of our protease inhibitor cocktails, such as Product No. P8340 for mammalian cells or Product No. P2714 for general use.
Can Product C2978 CelLytic™ M be used for adherent cells?
Yes, CelLytic™ M can be used for both adherent cells and cells in suspension. Use 125 μl CelLytic™ M for 106-107 of suspended cells. For adherent cells, use 500-1,000 μl for a 100 mm plate; 200-400 μl for a 35 mm plate.
