Biochem/physiol Actions

Inhibits release of catecholamines from adrenal medulla, acetylcholine at neuromuscular junctions.
Neurotoxin that inhibits release of catecholamines from adrenal medulla; blocks release of acetylcholine at neuromuscular junctions.

Physical form

Lyophilized powder containing 20 mM HEPES and 1.25% lactose when reconstituted with the volume of water or buffer indicated on the Certifcate of Analysis.

Preparation Note

Reconstitute each 100 μg vial with 100 μL of appropriate buffer. Upon reconstitution, the vial will contain 20 mM HEPES and 1.25% lactose. To insure full recovery of toxin from the vial, include 1 mg/mL BSA in the reconstitution buffer.
When toxin is used for in vitro studies, it is preincubated in a buffer containing 5 mM dithiothreitol for 30 minutes in order to reduce and thereby activate the toxin.

Other Notes

Handle the toxin gently. Do not vortex. Use sufficient volume of buffer to wash down the walls and stopper. Read all handling information found on SDS prior to reconstitution.

Giant liposomes were made from a mixture of asolectin phospholipid vesicles and presynaptic plasma membranes isolated from Torpedo cholinergic nerve endings. Acetylcholine filled giant liposomes were able to release neurotransmitter upon stimulation by the Ca2+ ionophore A23187 and Ca2+. Botulinum
