Application

HydroMatrix has been shown to support improved growth and migration in the proliferation of many cell types, including neural stem cells, neurons, glia, astrocytes, fibroblasts, and keratinocytes.

Components

HydroMatrix is a peptide nanofiber 3-dimensional scaffold that promotes cell growth and migration. It utilizes specific peptides that self-assemble from fluid precursors into highly crosslinked peptide hydrogels when presented with increased temperatures or ionic strengths. By inducing a rapid transformation from solution to gel, a peptide nanofiber scaffold is generated. Adjusting the concentration of the HydroMatrix solution, researchers can control the flexibility of the 3-D architecture to meet their application′s specific needs.

Caution

The lyophilized product at -20°C and is stable for many years at this temperature. The hydrogel should be stored at -70°C to maximize its active span. The 1% (w/v) stock solution should be stored -20°C and is stable for 3-6 months.

Preparation Note

This product is supplied as a lyophilized powder. A 1% (w/v) stock solution at 10 mg/mL can be prepared with addition of sterile water, resulting in a rigid gel. A 0.5% (w/v) solution will lead to a softer gel that is more suitable for cell migration and angiogenesis. Lower concentrations in the range of 0.15%-0.25% are more suitable for many cell types including neurons. In aqueous solution, HydroMatrix has a pH of 2.5 and should not be added directly to cells without prior addition of a buffer or medium. Once the hydrogel has formed, do not attempt to mix the gel any further, as this risks destroying the hydrogel integrity. Manipulation of the cells should be carefully performed to avoid disruption of the scaffold.

    The package size of the A6982 indicates the volume of H2O to use for reconstitution.  For example, a 1 ml package will require 1 ml of  H2O for reconstitution.

  6. Will Product A6982, HydroMatrix Peptide Cell Culture Scaffold, work will for adherent cells and suspension cells?   

    Yes, the product works with both cell types.

  7. What is the concentration of Product A6982, HydroMatrix Peptide Cell Culture Scaffold, when reconstituted as indicated?

    A 1% (w/v) (10 mg/ml) stock solution is accomplished by the addition of sterile dH20 as indicated.

  8. What concentration of HydroMatrix Peptide Scaffold, Product A6982, should I use for my cells?

    A 1% (w/v) solution results in a stronger gel. A working concentration of 0.5% (w/v) results in a soft gel with weak mechanical strength that is suitable for cell migration and angiogenesis.  Working solution concentrations as low as 0.15-0.25% are suitable for many cell types, including neurons. It is recommended for most applications to start with a 0.25% (w/v) working solution (2.5 mg/ml).

  9. How do I embed or encapsulate cells in HydroMatrix Peptide Scaffold, Product A6982, before polymerization?

    In a solution of dH20, HydroMatrix has a pH of 2.0-2.5; therefore it must not be added directly to cells.  The addition of buffer or media is performed prior to the addition of cells.  If performing cell encapsulation studies, addition of iso-osmotic sucrose will help protect the cells until a physiological pH is obtained.

  10. How does HydroMatrix Peptide Cell Culture Scaffold compare to PuraMatrix from Becton-Dickenson?

    Both products are peptide hydrogels.  There is no similarity between the peptides used to create the matrices.  They are both just classified as self-assembling peptide hydrogels. The sequence for the peptide is considered proprietary.
