General description

The Sephadex® ion exchangers contain a cross-linked dextran matrix with functional groups. These groups are linked by stable ether linkages to the glucose units in the matrix. DEAE Sephadex can be used in ion exchange chromatography for purifying and isolating proteins.

Application

DEAE-Sephadex® A-25 has been used:
  • to form DEAE-Sephadex A-25 (formate form) for glucosinolates (GLS) extraction using horseradish plants
  • in the estimation of GLS content in Chinese kale sprouts
  • production and purification of endozepine from astrocytes
  • to identify and label an insulin-activated nitric oxide synthase inhibitor (protein) in acute myocardial infarction (AMI) patients

    Unused media can be stored in the container at +4 to +30 °C.  Ensure that the screw-top is fully tightened.  Packed columns should be equilibrated in working buffer containing 20% ethanol to prevent microbial growth.

    According to the manufacturer, this is compatible with all commonly used aqueous buffers, 1 M NaOH, 8 M urea, 6 M guanidine hydrochloride and 70% ethanol.

    After packing, and before a chromatographic run, equilibrate with working buffer by washing with at least 5 bed volumes.

    After each separation, elute any reversibly bound material either with a high ionic strength solution (e.g. 1 M NaCl in buffer) or by increasing pH. Regenerate the media by washing with at least 5 bed volumes of buffer, or until the column effluent shows stable conductivity and pH values.
