General description

Phosphatase Inhibitor Cocktail 1 includes a mixture of inhibitors that will inhibit the L-isozymes of alkaline phosphatase as well as serine/threonine protein phosphatases such as PP1 and PP2A. This product contains microcystinLR, cantharidin, and (−)-p-bromotetramisole. Phosphatase Inhibitor Cocktail 1 has been replaced with P0044.

Application

Phosphatase Inhibitor Cocktail 1 has been used in nuclear protein isolation and electrophoretic mobility shift assay (EMSA) for catenin activation. It has also been used in western blot analysis.

Biochem/physiol Actions

Phosphatases are active regulators of a number of important biological processes. Inhibition of phosphatases by small molecules is useful to study its biological role.

Quantity

Recommended usage is a 100-fold dilution.

Analysis Note

Tested on cell extracts from animal tissues such as bovine liver, human placenta, and rabbit muscle, as well as on extracts from A431 and Jurkat cell lines.

  2. How stable is Product P2850, Phosphatase Inhibitor Cocktail 1?  Can I freeze it?

    This reagent, as supplied and stored at 2-8°C, will be stable for four years from the date of manufacturing. Freezing is not recommended, because it may cause precipitation of the inhibitors.

  3. At what concentration should I use Product P2850, Phosphatase Inhibitor Cocktail 1?

    In most circumstannces, the product is used at a 100-fold dilution.  This will inhibit the phosphatases in 5 mg of protein per mL of lysate.

  4. Product P2850, Phosphatase Inhibitor Cocktail 1, is solid when it arrived. Is it suitable to use?

    Since this solution is supplied in DMSO, it may solidify (melting point of DMSO is 16-19 deg C).  Simply allow it to melt before use.  This will not affect the product.

  5. What inhibitors are present in Product P2850, Phosphatase Inhibitor Cocktail 1, and in what concentrations?

    Cantharidin (Product Code C7632) inhibits protein phosphatase 2A (PP-2A).  Bromotetramisole (Product Code B8132) inhibits alkaline phosphatases. Microcystin LR (Product Code M2912) inhibits protein phosphatases 1 and 2A (PP-1 and PP-2A). The concentrations of these inhibitors are considered proprietary.

  6. Is Product P2850, Phosphatase Inhibitor Cocktail 1, suitable for use on yeast? 

    As far as we know, the phosphatases in mammalian cells also appear in yeast cells. Thus both cocktails, P5726 (Phosphatase cocktail II and P2850 (Phosphatase Inhibitor cocktail I), may be needed with yeast extracts.
