General description

Thioglycollate Medium is a powdered broth medium designed for the cultivation of both aerobic and anaerobic microorganisms. The medium contains dextrose, tryptone, yeast extract, and L-cystine providing essential growth factors for bacterial proliferation. Sodium thioglycollate and L-cystine serve as reducing agents that prevent the accumulation of lethal peroxides, enabling the growth of Clostridium even under aerobic conditions. Resazurin acts as an oxidation-reduction indicator, appearing pink when oxidized and colorless when reduced. The minimal amount of agar in the medium supports the growth of both aerobes and anaerobes, regardless of the presence of sodium thioglycollate. Sodium thioglycollate also neutralizes the toxic effects of mercurial preservatives and peroxides, promoting anaerobiosis and making the medium suitable for testing materials containing heavy metals. Any increase in oxygen content is indicated by a color change in resazurin from colorless to pink. The small agar content helps maintain a low redox potential, stabilizing the medium.

Application

Thioglycollate Medium is widely used in clinical testing, environmental monitoring, food and beverage testing, and sterility testing. It supports the growth of both aerobic and anaerobic microorganisms, making it ideal for sterility control in pharmaceutical products and for testing materials containing heavy metals with bacteriostatic effects. The medium is also used to determine an organism′s oxygen requirements and its ability to grow in anaerobic or microaerophilic environments.

Preparation Note

Solve 29.8 g in 1 litre distilled water. Autoclave 15 minutes at 121°C.
