General description

Schiff’s fuchsin-sulfite reagent is a part of the glycoprotein detection kit. It is primarily used in the Periodic Acid-Schiff (PAS) staining method to detect and visualize aldehydes, glycoproteins in biological samples. The reagent consists of basic fuchsin dye and sodium bisulfite. It reacts with the aldehyde groups of carbohydrates and produces a pink to magenta color, allowing for the identification of these substances in samples.

Application

Schiff′s fuchsin-sulfite reagent has been used:

  • in periodic acid-Schiff (PAS) staining to determine goblet cells and salivary glycopreoteins like mucins
  • to stain ovaries and testis
  • to stain tomato and Medicago primary roots
  • to stain undifferentiated sarcomas (USARCs)
It provides a system to easily detect the sugar moieties of glycoproteins on SDS PAGE or Western Blotting membranes.

Preparation Note

Prepared using pararosaniline.
