General description

Glycerol alters the hygroscopic property of gelatin and its permeability to water vapour. Glycerol Gelatin or glycerine jelly medium is an aqueous slide mounting media, used for mounting samples in microscopy and histology.

Application

Glycerol Gelatin has been used for mounting endometrial tissue and pancreatic sections for microscopic examination. It has also been used for the immobilization and mounting of helminth eggs for on chip image analysis.

Components

Prepared with gelatin, glycerol and phenol. Very similar in composition to Kaiser′s glycerol jelly.

Preparation Note

Product must be warmed to 55-60 °C before use. At ~37 °C the product returns to semi-solid state.

  1. How does the storage temperature relate to shipping conditions?

    The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.   6. Is Glycerol Gelatin (Product No. GG1) suitable for use with fluorescent dyes?

    Glycerol Gelatin is not recommended for use with fluorescent dyes.  Product numbers for suitable mounting medias for fluorescence would include 10979, 10981, F4680, F6182, F6057 and F5932.

  7. What is the refractive index of Glycerol Gelatin (Product No. GG1)?

    The refractive index of product GG1 is not measured on a lot to lot basis.  Expected refractive index is expected to be around 1.47.

  8. There are air bubbles in the Glycerol Gelatin (Product No. GG1).  How can these be removed?

    Once the air bubbles have formed in the Glycerol Gelatin, the air bubbles are most easily removed.  The preferred method is to stop the formation of air bubbles.  To use the GG1, the bottle is turned upside down during use.  If the bottle is kept upside down while being utilized, no or fewer air bubbles will form.  Air bubbles will form when the bottle is constantly turned upside down and then righted again for each 1-2 slides.

  9. Is Glycerol Gelatin (Product No. GG1) a suitable mounting media for hematoxylin and eosin (H&E) stained slides?

    Glycerol Gelatin is an aqueous mounting media and not recommended for use in coverslipping H&E stained slides.  Fading would occur more rapidly, as compared to standard permanent mounting medias - such as DPX Mountant for histology.

  10. How long before slides coverslipped with Glycerol Gelatin (Product No. GG1) begin to fade?

    Fading will occur when slides are coverslipped with Glycerol Gelatin. The amount of time will vary by dye and application. Fading could occur in as few as a few days or after a period of several months. If a long term record of the staining is desired, the user should consider photographing the stained slides.
